Uji Kepekaan terhadap Antibiotik dan Kemampuan Membentuk Biofilm pada Escherichia Coli Penyebab Infeksi Saluran Kemih
Abstract
Escherichia coli merupakan bakteri yang sering menyebabkan infeksi saluran kemih (ISK) dan memiliki kemampuan membentuk biofilm di dalam jaringan, sehingga mengakhibatkan pengobatan infeksi menjadi lebih sulit. Tujuan Penelitian ini adalah mengetahui kepekaan E. coli yang mengakibatkan infeksi saluran kemih terhadap beberapa antibiotik dan kemampuannya dalam membentuk biofilm. Penelitian ini didahului dengan identifikasi E. coli yang diisolasi dari sampel urin penderita ISK dengan metode uji biokimia dan uji kepekaan berbagai antibiotik dengan metode dilusi (vitek 2 Compact). Uji kemampuan pembentukan biofilm dilakukan dengan metode Microtiter Plate Culture pada media Trypticase broth, diinkubasi selama 24 jam dengan suhu 37oC. Biofilm yang terbentuk diukur berdasarkan masa yang melekat pada sumuran micotiter plate 96 well dengan pewarnaan crystal violet 0,1%, dibaca dengan microplate reader pada panjang gelombang 570nm, dengan masing-masing ulangan 12 kali. Pengukuran biofilm berupa nilai Optical density dikategorikan dalam kuat, sedang, dan lemah. Hasil penelitian menunjukan semua isolat E. coli memiliki kesamaan sifat biokimia, namun memiliki kepekaan yang bervariasi terhadap beberapa antibiotik. Ada 14% isolat memiliki kemampuan pembentukan biofilm kuat, dan 43% isolat sedang, dan 43% isolat lemah, tidak ditemukan hubungan antara pembentukan biofilm dan kepekaannya terhadap antibiotik.
