Aktivitas Sitotoksik, Antioksidan dan Adipogenesis Hidrolisat Kolagen dari Ceker Ayam

Abstract

Saat ini orang lebih suka menggunakan produk alami karena keamanan kesehatannya. Eksplorasi produk alami dan manfaatnya terus dikembangkan sebagai pangan fungsional dan kemungkinan memberikan penyembuhan tanpa efek samping. Salah satunya adalah eksplorasi betis ayam sebagai sumber kolagen dan peptida sebagai sitotoksik, antioksidan dan adipogenesis. Penelitian ini dibagi menjadi 3 tahap: 1. Ekstraksi kolagen, 2. Hidrolisis kolagen, 3. Uji bioaktivitas meliputi sitotoksik, antioksidan dan adipogenesis. Ekstraksi kolagen menghasilkan rendemen 9,48% dengan berat molekul 130 kDa. Hidrolisis kolagen dengan enzim kolagenase menghasilkan derajat hidrolisis tertinggi pada konsentrasi enzim optimum 5%, pH 8, suhu 50 derajat celcius, waktu inkubasi 45 menit dengan persentase 87,53%. Hasil penelitian menunjukkan bahwa hidrolisat kolagen memiliki sitotoksik, antioksidan dan adipogenesis yang lebih baik daripada kolagen. Toksisitas HepG2 pada kolagen hidrolisat adalah IC50 347 µg / mL sedangkan kolagennya adalah IC50 1410µg / mL. Toksisitas pada sel kanker payudara MCF7 kolagen hidrolisat adalah IC50 958 µg / mL sedangkan kolagen adalah IC501626 µg / mL. Aktivitas pembersihan radikal (diukur dengan DPPH) dari kolagen hidrolisat lebih tinggi (7,1%) dibandingkan dengan kolagen (2,3%). Uji akumulasi lipid kolagen hidrolisat tertinggi (299%) sedangkan kolagen 229%.
